Permissions can cause problems if they aren't correct.In IE you can open Tools - Internet Options - Advanced Options and uncheck the "show friendly http errors" setting. Often this will give you enough detail on your error to identify the problem.

you can see the tree doesn't mean it is started. in command window, run "iisreset" to restart your iis server and make sure the state of your default web site is running, not stopped

Try this:From your IE, click Tools - Internet Options,Switch to Connections tab,
Click on "LAN Settings" button at the bottom.If "Automatically detect settings" is checked in the next section uncheck it, then OK and Apply.
Close your IE and restart it.
Try it again.

Did you try this?In your .NET Framework runtime directory (typically C:\Windows\Microsoft.NET\Framework\v1.0.3705 or later version ), run:
aspnet_regiis.exe -i
If you really cannot find the reason, usually uninstall and reinstall iis would be helpful. Uninstall and reinstall iis, try the command above again. but you have to rebuild your virtual directory yourself.

500 error is friendly error from miscrosoft, disabling friendly error in your browser will bring you exact technical error generated from your code. got it?No? send me your email, I will tell you step by step :-)
